Replenishment of the genetic bank of endemics, rare and economically valuable plants(seed bank and collections of tissue cultures and cultures of isolated organs - roots) and development of methods for the conservation and rational use of plant resources

Section 1. Replenishment of the seed bank of plants.

  Work continues on the project “Banking seeds of the flora of Kyrgyzstan 2016-2020” with the Royal Botanic Gardens, Kew. Despite of pandemic COVID-19, the laboratory staff conducted expeditions to collect seeds and herbarium material. To date, work has been completed to prepare seeds for long-term storage at low temperature conditions in the seed bank of the Institute of Biotechnology and send duplicate material to the Royal Botanic Gardens, Kew.

In 2020, seeds of 142 plant species were collected. For our bank, we left the seeds of 74 species of plants, the seeds of 58 species were new to our bank. Of these 58 species, 36 are widespread, 13 are subendemic and 4 are endemic.

In the winter of 2020-2021, the seeds collected in 2020 were prepared for storage.

The seeds were cleaned, the weight of the seeds and their number were determined.

The seeds were dried to the optimum moisture content and stored in a bank.

 Seeds of 75 species of plants were prepared and sent to MSB, seeds of 51 species are new for MSB.

Thus, we have collected and stored seeds of approximately 30% of plant species in Kyrgyzstan.

In April, May and June 2021, our laboratory made several trips around the Chui Valley to collect seeds of ephemeroid plants.

 In July, we worked to collect material in the gorges in the Jalal-Abad region and the Sary-Chelek nature reserve.

In August, seeds were collected in the Chatkal valley.

During these expeditions, seeds of 81 plant species from 28 families were collected.

 Seeds of plant species will be prepared and sent to MSB

Seeds of ornamental species were specially collected for the MSB project to create landscapes of the Great Silk Road.

In our constantly maintained collection there are 3 species of rare potentially useful plants of Kyrgyzstan of the genus Scutellaria,

 regular transplants of these plants are carried out (every 1.5 months)

Callus cultures

Callus cultures of the following types are supported in liquid culture and on agar medium

Scutellaria botbaevae Lazkov

The callus of Scutellaria andrachnoides Vved is being cultivated. to obtain quantities of lyophilized material sufficient to determine the biological activity. Constant monitoring of the content of the main flavonoids in the collected material is carried out.

 In our collection there are 3 species of rare potentially useful plants of Kyrgyzstan of the genus Scutellaria. Regular transplants of these plants are carried out (every 1.5 months)

In the current year, we have supported and carried out micropropagation of previously obtained germplasm lines of Scutellaria andrachnoides species in order to obtain plant material for biochemical studies.

Transformed Roots

1) Permanent transplants of 4 lines of transformed Sc roots are carried out. andrachnoides obtained in previous years. These cultures are grown both on agar medium and in liquid shake culture. There is a collection of material to determine the biological activity.

2) Maintained in liquid culture and on agar medium callus culture Sc. andrachnoides, which was transferred to us from the Group of Specialized Root

Metabolism of the IGF RAS.

The callus is cultured to obtain quantities sufficient to determine biological activity. Constant monitoring of the content of the main flavonoids in the collected material is carried out.

3) Transplantation is being carried out and the material of two selected lines of culture of the transformed roots of Scutellaria przewalskii is being accumulated.
